Acid and enzymatic hydrolysis to recover reducing sugars from cassava bagasse: an economic study BABT

The objective of this work was to study the acid and enzymatic hydrolysis of cassava bagasse for the recovery of reducing sugars and to establish the operational costs. A statistical program "Statistica", based on the surface response was used to optimize the recovery of reducing sugars in both the processes. The process economics was determined considering the values of reducing sugars obtained at laboratory scale, and the operations costs of a cylindrical reactor of 1500 L, with flat walls at the top and bottom. The reactor was operated with 150 kg of cassava bagasse and 1350 kg of water. The yield of the acid hydrolysis was 62.4 g of reducing sugars from 100 g of cassava bagasse containing 66% starch. Alkali pretreated of wheat straw and its enzymatic hydrolysis BJM

The efficiency of enzymatic hydrolysis of cellulose can be improved by various pretreatments of the substrate. In order to increase the efficiency of enzymatic saccharification of the wheat straw, we determined the effect of different pretreatments on the physical structure, chemical components and enzymatic saccharification of wheat straw. Our results showed that combination of grinding and sodium hydroxide (NaOH) treatment had high effect on the enzymatic hydrolysis of wheat straws. The optimal pretreatment condition was to grind the wheat straws into the sizes of 120 meshes followed by treatment with 1.0% NaOH for 1.5 h (121°C/15psi). Anaerobic biodegradability of dairy wastewater pretreated with porcine pancreas lipase BABT

Lipids-rich wastewater was partial hydrolyzed with porcine pancreas lipase and the efficiency of the enzymatic pretreatment was verified by the comparative biodegradability tests (crude and treated wastewater). Alternatively, simultaneous run was carried out in which hydrolysis and digestion was performed in the same reactor. Wastewater from dairy industries and low cost lipase preparation at two concentrations (0.05 and 0.5% w.v-1) were used. All the samples pretreated with enzyme showed a positive effect on organic matter removal (Chemical Oxygen Demand-COD) and formation of methane. Resumo: Um dos principais desafios da coleta e extração de óleo de frutos de dendê (palma de óleo) é a manutenção da qualidade do óleo, de forma a evitar a liberação de ácidos graxos, que mudam as propriedades do óleo e, consequentemente, reduzem seu valor comercial. A liberação desses ácidos é atribuída à ação de lipases. Entretanto, não há um consenso quanto à fonte das enzimas que hidrolisam excessivamente o óleo, se ela é endógena (produzida pelo próprio fruto de dendê) ou exógena (produzida por um determinado microrganismo que colonizou o fruto de dendê). Dans le présent travail, l’effet du séchage solaire sur la composition biochimique et la qualité des lipides des athérines a été étudié. La teneur en lipides, protéines et en cendres dans l’athérine a augmenté significativement (p <0.05) après le séchage. Cependant, celle de l’humidité a montré une forte diminution pour atteindre 22.37% dans le produit final. D’autre part, l’étude des indicateurs d’altération des lipides (Oxydation et Hydrolyse) nous a montré que la qualité des lipides des athérines est affectée par ce type de traitement. The aim of this study was to identify the most adequate level of calcium oxide (CaO) in the treatment of sugar cane by evaluating the chemical composition and in vitro digestibility of dry matter. The sugar cane was homogenized with CaO levels 0, 0.75, 1.5, 2.25, 3.0, 3.75 and 4.5%, in natura matter, for 24 hours. The dry matter and mineral matter increased, while the organic matter of the sugar cane decreased (P <0.05) linearly as a result of the CaO levels. The CaO had no effect on the levels of crude protein and ether extract. The phosphorus concentration remained unchanged, but there was a significant increase (P<0.05) in the calcium content. Fish sauces are traditionally used in most Asian countries. Whatever the process, the principle is the same i.e. a fish autolysis in saline media followed by a fermentation step. Not less than 18 months are needed to obtain a brown liquid rich in salt and amino acids. This PhD work aims to determine if an addition of proteases can speed up the process while keeping the nutritional and sensorial properties. Degradation of Technora aramid fibres in alkaline and neutral environments ArchiMer

Technora fibres are high performance aramid fibres which have been used in ropes and protective clothing for many years. They are also now being proposed as geotextiles for soil reinforcement. However, there is a lack of experience on the long-term behaviour of Technora fibres in an alkaline environment (lime-treated ground.). Consequently, aging studies have been performed under different conditions (deionised water, pH9 and pH11). Hydrolytic degradation has been evaluated by FTIR, viscometry, TGA, density measurements, SEM, and by tensile measurements. Some chain degradation and finish rearrangements have been highlighted, but Technora fibres retain their mechanical properties for all the conditions considered here.

Effect of intraperitoneally administered hydrolyzed whey protein on blood pressure and renal sodium handling in awake spontaneously hypertensive rats BJMBR

The present study evaluated the acute effect of the intraperitoneal (ip) administration of a whey protein hydrolysate (WPH) on systolic arterial blood pressure (SBP) and renal sodium handling by conscious spontaneously hypertensive rats (SHR). The ip administration of WPH in a volume of 1 ml dose-dependently lowered the SBP in SHR 2 h after administration at doses of 0.5 g/kg (0.15 M NaCl: 188.5 ± 9.3 mmHg vs WPH: 176.6 ± 4.9 mmHg, N = 8, P = 0.001) and 1.0 g/kg (0.15 M NaCl: 188.5 ± 9.3 mmHg vs WPH: 163.8 ± 5.9 mmHg, N = 8, P = 0.0018). Effect on the enzymatic hydrolysis of lipids from dairy wastewater by replacing Gum Arabic emulsifier for sodium chloride BABT

The objective of this work was to evaluate the replacement of Gum Arabic for sodium chloride to reduce fat and organic contents in dairy wastewater using two low cost commercially available lipase preparations from animal source (Kin Master - LKM and Nuclear- LNU). The best performance was achieved when lipase Nuclear (LNU) was used as catalyst. In addition, this lipase preparation has also lower cost, which makes its use a quite promising technique for reduction of suspended solids as proteins and lipids contents found in wastewater generated by dairy industries.

Enzymatic hydrolysis of sugarcane bagasse pretreated with acid or alkali BABT

The aim of this study was to evaluate the performance of enzymatic hydrolysis of acid or alkali pretreated sugarcane bagasse for the production of fermentable sugars. The first step consisted of selection of commercial enzymes presenting the highest cellulolytic activities. After selection of four enzymes: HPL, CL, P1 and P4, their performances were tested in the bagasse pretreated with acid and alkali. The sugar content of the hydrolysates was analyzed by anion exchange liquid chromatography. Data showed that the joint action of 0.5% acid pretreatment, 121ºC, 30 minutes and enzyme CL provides the best results, 67.25 g of hexose and 148.13g of pentose per kg of dry bagasse.

Extracellular amylase production of a thermotolerant Fusarium sp: isolated from Eastern Nigerian soil BABT

In this work, an α- amylase producing Fusarium sp. was isolated from the soil at 50 0C. Growth and enzyme production occurred at 30, 45 and 55 ºC. Soybean meal at 1% concentration, supplemented with 0.2% NH4Cl and 2.5% corn starch elicited the highest amylase yield. Optimum pH for the enzyme was pH 6.5 which retained over 60% of its activity after 24 h incubation at the pH range of 4.5-7.0. The enzyme showed high activity from 40-70 0C with optimal activity at 50 ºC and 78% activity was retained after incubation at 70 ºC for 30min. Catalytic function of the crude amylase was stimulated by Mg (136%), Ca (118%) and Zn (118%) at 2mM concentrations. The enzyme hydrolyzed cassava, potato and yam starches effectively.

Hydrolysis of insoluble fish protein residue from whitemouth croaker (Micropogonias furnieri) by fungi BABT

A significant amount of insoluble fibrous protein, in the form of feather, hair, scales, skin and others are available as co-products of agro industrial processing. These wastes are rich in keratin and collagen. This study evaluated different fungi for the hydrolysis of insoluble fish protein residues. Proteins resulting from Micropogonias furnieri wastes through pH-shifting process were dried and milled for fermentation for 96 h. This resulted the production of keratinolytic enzymes in the medium. Trichoderma sp. on alkaline substrate (28.99 U mL-1) and Penicillium sp. on acidic substrate (31.20 U mL-1) showed the highest proteolytic activities.
